We have used an antibody raised against a peptide from the extracellular domain of the 180 kDa SCP2 (SCP2-ab) to test for possible sterol binding, and to examine for sterol binding activity in the PM vesicles. SCP2-ab binding was found to be sigmoidal in concentration and was saturable at a molar ratio of 2:1. Molar ratios of 10:1, 20:1 and 60:1 of sterol to SCP2-ab caused significant and dose-dependent inhibition of SCP2-ab binding, suggesting that the sterol binding site in SCP2-ab is hydrophobic in nature and the binding site is greater than 25 A from the protein surface.
