We have investigated the optimum concentration of 5-aminolevulinic acid (ALA) for hematoporphyrin production in a rat tumor model by continuous administration of ALA into the small intestine. While a single oral administration of 930 mg/kg of ALA produced 8.2 mg/g of hematoporphyrin at 2 hours after administration in the solid tumor-bearing rat, continuous administration of 10-20 mg/kg of ALA for 6 hours produced 10-50 times as much hematoporphyrin as this dose. When the maximum tolerated dose (MTD) of ALA was examined in the tumor-bearing animal, a dose of 590 mg/kg of ALA was found to be the MTD, showing that 1/4 of the MTD is the optimum concentration of ALA for hematoporphyrin production. There was no significant difference in the efficacy and dosage-response of hematoporphyrin production between the solid tumor and the ascites tumor. The tryptic peptide mapping of the haem-dependent peroxidase (HDP) isolated from the fungus Talaromyces stipitatus was studied by circular dichroism (CD). In the far-UV CD region, a regular secondary structure with three alpha-helices and three beta-strands forming a jelly-roll like structure was observed. The alpha-helices are both parallel and antiparallel in beta-sheet direction. The CD spectrum at wavelength regions of 250 nm and 280 nm shows a clear maximum at wavelength of 250 nm. The simultaneous occurrence of positive and negative ellipticity changes implies that the intensity of the CD signal depends on the orientation of the chromophore within the protein (i.e. the spatial relationship of the tryptophan and haem in solution). In the near-UV region, the fluorescence emission maximum was observed at 347 nm. Based on this wavelength, we have calculated the emission spectrum of the haem environment (lambda(ex) = 340 nm and lambda(em) = 390 nm). The results point to a medium-polar environment for the ligand. A tertiary structure model was made using a combination of the known topology of HDP from Escherichia coli and known topologies of plant HDPs.
